The Tsokos laboratory studies immune cell signaling, metabolism and gene transcription in patients with systemic autoimmune diseases using state-of-the-art approaches. Our aim is to identify molecules that are central to the immunopathogenesis of the disease and can be exploited therapeutically in the clinic. In parallel, disease biomarkers and approaches to provide personalized medicine are been developed. Studies in humans are expanded in normal and lupus-prone mice in which identified molecules are overexpressed or deleted globally or conditionally. Animals are used to provide further mechanistic insights and sufficient preclinical rationale for clinical trials. The laboratory has developed a strong interest in the interface of the immune system with tissue resident cells and in the identification of local factors that enable the development of organ injury. Approaches to develop cell-targeted treatment modalities are being developed.
